🎯 Workshop Objectives
General: Train participants on Oxford Nanopore Technologies MinION Mk1D and PromethION 2 Solo using the Rapid Barcoding Sequencing Kit 24 V14 (SQK-RBK114.24) or Ligation Sequencing Kit DNA V14 (SQK-LSK114.24).
Specific:
- Train end-users on the end-to-end workflow of Rapid Barcoding or Ligation Sequencing.
- Train end-users on the operation of MinION Mk1D and PromethION 2 Solo devices.
- Discuss troubleshooting and best practices for Nanopore sequencing.
📋Pre-Training Preparation
Submission of attendee names prior to the workshop for Training Certificate generation.
🔬Methodology
The workshop opens with an overview of the Rapid Barcoding or Ligation Sequencing workflow, followed by presentation-supported guidance and hands-on sample quality checks, hardware & flow cell checks, flow cell priming, and library loading. Participants execute a hands-on sequencing run with trainer support and interactive Q&A throughout.

- 200ng gDNA / sample (or Lambda)
- Rapid Barcoding Kit 24 V14 / Ligation Kit DNA V14
Consumables:
- MinION & PromethION Flow Cells R10.4.1
- 1.5mL / 2.0mL LoBind tubes & 0.2mL PCR tubes
- Nuclease-free water, 100% Ethanol, BSA
- Qubit dsDNA Kit & wide bore pipette tips
Equipment:
- MinION & PromethION 2 Solo Devices
- Qubit Fluorometer & Magnetic Rack
- Thermal cycler, Rotator mixer, Pipettes
- Ice bucket, Vortex, Minicentrifuge, Timer
🎓Expected Output
The trainees are expected to have learned the whole Rapid Barcoding or Ligation Sequencing workflow; as well as the best practices and troubleshooting of Nanopore Sequencing.
